David Baltimore was born on March 7, 1938, in New York City to Gertrude (Lipschitz) Baltimore and Richard Baltimore. He grew up in Queens (Forest Hills and Rego Park) before the family moved to suburban Great Neck so he could attend stronger schools. His father had been raised Orthodox Jewish; his mother was an atheist who later earned advanced degrees and became a tenured psychology professor at Sarah Lawrence College. Baltimore observed Jewish holidays with his father through his Bar Mitzvah. A high-school summer at the Jackson Laboratory in Bar Harbor, Maine, fixed his vocation in research biology. There he met Howard Temin, with whom he would later share the Nobel Prize.
He earned a B.A. with high honors in chemistry from Swarthmore College in 1960, after switching from biology so he could complete a research thesis. A 1959 Cold Spring Harbor summer with George Streisinger deepened his commitment to molecular biology and connected him to Salvador Luria and Cyrus Levinthal, who recruited him toward MIT. He began graduate work at MIT in 1960, then shifted to animal virology, taking the Cold Spring Harbor animal virus course and completing his Ph.D. at Rockefeller University in 1964 with Richard Franklin on mengovirus and macromolecular synthesis. Postdoctoral work followed with James Darnell at MIT and Jerard Hurwitz at Albert Einstein College of Medicine.
In 1965 Renato Dulbecco recruited him to the new Salk Institute in La Jolla. There he worked on poliovirus RNA replication and proteolytic processing of viral polyproteins, and met virologist Alice S. Huang, who joined his orbit in 1967. In 1968 he moved to MIT as associate professor of microbiology; Huang moved with the VSV project. They married in October 1968. At MIT, work on vesicular stomatitis virus polymerase set up his decisive 1970 experiment: demonstrating RNA-dependent DNA polymerase (reverse transcriptase) in RNA tumor viruses, published back-to-back in Nature with Temin and Satoshi Mizutani. The finding overturned the simplified central dogma that genetic information flows only DNA to RNA to protein, defined retroviruses (including later HIV), and became a cornerstone of molecular cloning and biotechnology. In 1971 he also published the Baltimore classification of viruses by genome and mRNA pathway. In 1975, at age 37, he shared the Nobel Prize in Physiology or Medicine with Temin and Dulbecco.
The same year he helped Paul Berg, Maxine Singer, and others organize the Asilomar Conference on Recombinant DNA, shaping voluntary biosafety guidelines that influenced NIH oversight worldwide. He later co-chaired the 1986 National Academy of Sciences / Institute of Medicine Confronting AIDS report calling for a major federal research program, and in 1996 headed the NIH AIDS Vaccine Research Committee. His MIT and Whitehead labs produced landmark immunology: NF-κB (with Ranjan Sen, 1986), RAG-1/RAG-2 recombination genes (Schatz and Oettinger), Abelson virus tyrosine kinase work that fed into Gleevec's scientific lineage, and infectious poliovirus clone work with Vincent Racaniello.
In 1982 he became founding director of the Whitehead Institute for Biomedical Research at MIT, building a model independent institute and the Whitehead Fellows program. He served as Rockefeller University president (1990-1991), resigning amid the Thereza Imanishi-Kari controversy (he was never charged with misconduct; Imanishi-Kari was later cleared on appeal in 1996). He returned to MIT, then became Caltech's president (1997-2006), elevating life sciences, fundraising, and student life while keeping an active lab. After the presidency he remained at Caltech as professor (later Distinguished / Judge Shirley Hufstedler Professor of Biology), pursuing gene therapy for HIV (Engineering Immunity, Gates Grand Challenges), microRNA in immunity, and splicing control of inflammation until announcing lab closure around 2018-2019. He served as AAAS president (2007-2008 term; often cited as 2008), received the National Medal of Science (1999), and the Lasker-Koshland Special Achievement Award (2021).
Baltimore married Alice S. Huang in 1968. They had one daughter, Lauren Rachel Baltimore, known as TK (pronounced Teak), who married Jay John Konopka in 2001. Sources also mention a granddaughter. He was an avid fly-fisher and, friends said, a host with wide cultural appetites. He died of cancer at home in Woods Hole, Massachusetts, on September 6, 2025, aged 87.
